Abstract
Polarization-sensitive stimulated Raman scattering (SRS) microscopy allows noninvasive, fast, and background-free vibrational imaging of highly ordered and oriented molecules. In this chapter, we describe the principle of polarization-sensitive SRS microscopy, the setup, recent applications, and its use for resolving amphotericin B orientation in fungal cell membrane. Polarization-sensitive SRS microscopy offers an advanced approach to study molecule orientation and assembly at submicron spatial resolution, which opens new opportunities for disease diagnosis and drug development.
